Design Notes

1. Is there a simple method to detect the expression of repressor protein?Thus the GFP gene is retained to detect the expression level of lacI protein. 2. When linking gene fragments, homologous arms need to be designed on primers.


Source

The light-controlled genes such as J23100 promoter, yf1 protein and FixJ protein come from the plasmid dusk-egfp-puc57 stored in E. coli from 2018-zjut, while the repression system of lactose operon come from pET28b.
